Drug and dietary modification of platelet function

Abstract

A st u d y of pla t e l e c aggregation and MDA production after an oral dose of 300mg aspirin indicated that partial rec o v e r y of platelet function occ u r r e d when a p p r o x i m a t e l y one third of the c i r c u lating platelets had been rep l a c e d by new (uninhibited) platelets. In vitro studies on mixtures of normal and aspirin inhibited p l a t elets indicated partial restoration of platelet agg r e g a t i o n and thromboxane production w i t h as little as 10% of normal p l a t e l e t s in some subjects. Re s t o r a t i o n of full function required a higher proportion of normal platelets. There was considerable variation between subjects. These data suggest that complete suppression of p latelet functions in all normal subjects r equires d a i l y a d m i n i s t r a t i o n of the d r u g . Despite the h y p e r a g g r e g & b i l i t y of platelets in Familial H y p e r c h o l e s t e r o l a e m i a / ISO mg aspi r i n was shown to inhibit p l a t e l e t aggregation to the same e.ttent as occurs in normals.
